abstract = {Recurrent episodes of moderate to severe headache accompanied by sensory and autonomic symptoms are the hallmarks of migraine, a common and incapacitating neurovascular condition. Calcitonin gene-related peptide (CGRP) is a crucial mediator in trigeminovascular activation, vasodilation, neurogenic inflammation, and nociceptive transmission, according to recent developments in migraine research. CGRP is a 37-amino acid neuropeptide that is extensively produced in trigeminal sensory neurons and is essential for both peripheral and central sensitization processes. Increased neuronal excitability and vascular responses result from the activation of intracellular signalling pathways involving cyclic adenosine monophosphate by the CGRP receptor, which is composed of a calcitonin receptor-like receptor and receptor activity-modifying protein 1. CGRP is released when the trigeminovascular system is activated, and both peripheral processes, such as vasodilation, contribute to the onset and progression of migraines. CGRP, which is released when the trigeminovascular system is activated, has a role in the onset and progression of migraines through both peripheral processes like vasodilation and inflammation and central mechanisms like amplification of pain signaling.For the treatment of migraines, targeting CGRP has become a cutting-edge, mechanism-based approach. These pathways are successfully blocked by CGRP receptor antagonists and monoclonal antibodies that target CGRP or its receptor, which reduces migraine frequency, intensity, and related impairment. These medicines have higher specificity, greater tolerability, and a lower risk of systemic side effects than traditional therapies.},
