Development and Evaluation of sustained release empagliflozin matrix tablets using hydroxypropyl methylcellulose and guar gum

  • The present research work focuses on the development and evaluation of sustained- release matrix tablets of empagliflozin using hydroxypropyl methylcellulose ( HPMC) and guar gum as releaseretarding polymers. Empagliflozin is an oral antidiabetic drug used in the management of type 2 diabetics mellitus. Conventional dosage forms require frequent administration and may produce fluctuations in plasma drug concentration. Sustained-release formulations are designed to maintain therapeutic drug levels for an extended period, improve patient compliance, and reduce dosing frequency. In this study, matrix tablets of empagliflozin were prepared by the direct compression method using varying concentrations of HPMC and guar gum. The prepared formulations were evaluated for precompression parameters such as angle of repose, bulk density, cars index, and Hausner ratio to determine the flow properties of the powder blend. Post-compression evaluation included thickness, hardness, friability, weight variation, drug content uniformity, and in-vitro dissolution studies. The dissolution studies were carried out using suitable dissolution media to assess the sustained-release behavior of the formulations. The results indicated that polymer concentration significantly affected the drug release profile. Formulations containing a combination of HPMC and guar gum showed controlled and prolonged drug release over an extended period. Stability studies demonstrated that the optimized formulation remained stable under specified storage conditions. The study concluded that sustained-release matrix tablets of empagliflozin can be successfully formulated using HPMC and guar gum, providing effective controlled drug release and improved therapeutic efficacy for the treatment of diabetes mellitus.
