Method Development and Validation of Simultaneous Estimation of Buprenorphine and Naloxone in Bulk and Pharmaceutical Dosage Forms by RP-HPLC Method

  • A rapid, sensitive, and precise reverse-phase high-performance liquid chromatography (RP-HPLC) method was developed for the simultaneous quantification of Buprenorphine and Naloxone using a Waters HPLC system. Chromatographic separation was achieved on an Inertsil ODS C18 column (250 × 4.6 mm, 5 µm particle size) maintained at ambient temperature. The mobile phase consisted of methanol and acetonitrile in the ratio of 85:15 (v/v), which was filtered through a 0.45 µm membrane filter prior to use. The flow rate was maintained at 1.0 mL/min, and detection was carried out at 247 nm using the PDA detector.
