Formulation and Evaluation of ibuprofen tablet

  • Ibuprofen is one of the most used and best-tolerated NSAIDs. Several preformulation studies have been performed to improve its bioavailability and obtain a more controlled release. In these studies, different formulations containing ibuprofen were prepared with two kinds of excipients, HPMC and microcrystalline cellulose (Avicel®), at different concentrations. The best formulation provided a steady state drug release rate close to 60 mg/hour. Other studies involved the investigation of granule particle size on dissolution behavior and the use of two different crystal forms of ibuprofen. Both crystal forms presented different release mechanisms from that obtained for the original crystalline form. Moreover, the effect of some surfactants like Tween 80 and SLS on drug release was also studied. All formulations containing SLS presented a faster disintegration of tablets compared to those containing Tween 80. This fact suggests that excipient selection is an important factor in order to optimize ibuprofen release profiles.
