  • Abstract:
  • A novel and simple Reverse phase liquid chromatographic method was established for the determination of Agomelatine in presence of degradants. Agomelatine is a potent agonist at melatonin receptors and an antagonist at serotonin-2C (5-HT2C) receptors, tested in an animal model of depression. It is used for the treatment of major depressive disorder. The proposed work was performed on Thermo Hypersil extended C18 column (250 mm × 4.6 mm i.e., 5 µm particle size) using a mixture of Buffer (4.3pH) and methanol (60:40) as mobile phase with flow rate 1.0 ml/min. Water and Methanol (50:50) was used as diluent. The retention time was found to be 3.374 min. The linearity range used was 25-75µg/ml. The %RSD was found to be less than 2%. LOD & LOQ values were found to be 3.3002 and 10.0008µg/ml respectively.
